The interaction of 7,12-dimethylbenz(a)anthracene with cells sensitive and resistant to toxicity induced by this carcinogen.

چکیده
The multiplication of monolayer cultures of transformed rodent cells and normal or transformed human cells was not inhibited by concentrations of 7,12-dimethyIbenz(a)anthracene (DMBA) 200-500 times greater than those that inhibited the multiplication of normal embryonic rodent cells. Fluorescence microscopy showed that DMBA localized to about the same degree within the cytoplasm of cells that were sensitive or resistant to the toxic effect of the carcinogen. Autoradiography with DMBA-3H and an aqueous cell fixative (formalin) revealed that the carcinogen was distributed throughout the cytoplasm and nucleus and that the concentrations were similar in sensitive and resistant cells. However, when the cells were treated with a fixative in which DMBA is soluble (Carnoy's), most (90%) of

Protective effect of dehydroepiandrosterone against aflatoxin B1-and 7,12-dimethylbenz(a)anthracene-induced cytotoxicity and transformation in cultured cells.
Dehydroepiandrosterone, an adrenal steroid that has been reported to be produced in subnormal amounts and found in lower plasma concentrations in women with benign and malignant breast tumors, protects cultured rat liver epithelial-like cells against aflatoxin B1-induced cytotoxicity and protects hamster embryonic fibroblasts against aflatoxin B1- and 7,12-dimethylbenz(a)anthracene-induced tran...
متن کاملBiochemical basis for cytotoxicity of 7,12-dimethylbenz(a)anthracene in rat liver epithelial cells.
When the effects of 7,12-dimethylbenz(a)anthracene (DMBA) on normal and malignant rat liver epithelial cells were compared in a colony inhibition assay, this carcinogen showed a preferential cytotoxic action on the normal cells. In investigations of the biochemical basis of this selective toxicity, it was found that both cell lines were similarly effective in binding DMBA to DNA and that both c...
متن کاملFlavonol-stimulated efflux of 7,12-dimethylbenz(a)anthracene in multidrug-resistant breast cancer cells.
We used a series of P-glycoprotein (P-gp) expressing multidrug-resistant (MDR) cells, developed from human breast cancer MCF-7 cells by exposure to Adriamycin, to investigate the effects of flavonoids on P-gp-mediated efflux mechanisms for chemical carcinogens. We previously showed that MDR cells derived from exposure to Adriamycin are cross-resistant to a chemical carcinogen, benzo(a)pyrene, d...
متن کاملRole of erythropoietin in 7,12-dimethylbenz(a)anthracene induction of acute chromosome aberration and leukemia in the rat.
The incidence of chromosome aberrations in rat bone marrow, examined 6 hr after the administration of 7,12-dimethylbenz(a)anthracene, was significantly enhanced by induction of anemia 0-48 hr before the carcinogen treatment and was suppressed by induction of polycythemia. The suppressive influence of polycythemia was reversed by sheep erythropoietin injected shortly before or after the carcinog...
متن کاملPotent tumor-initiating activity of the 3,4-dihydrodiol of 7,12-dimethylbenz(a)anthracene in mouse skin.
The abilities of the racemic trans-3,4-, 5,6-, and 8,9-dihydrodiols of 7,12-dimethylbenz(a)anthracene to initiate skin tumors in mice were determined by using a two-stage system of tumorigenesis. The 7,12-dimethylbenz(a)anthracene trans-3,4-dihydrodiol was found to be much more active as a tumor initiator than the parent hydrocarbon. The 7,12-dimethylbenz(a)anthracene trans-5,6- and 8,9-dihydro...
